Reaction Buffers for OPTIZYME™ Restriction Enzymes

Popularly known today as “Good” buffers, these organic buffers have several advantages compared to traditional inorganic buffering systems such as phosphate, borate or bicarbonate.

“Good” buffers approach the “ideal” buffer state by having pKa values at or near physiological pH and minimal interference with biological processes. JustPURE “Good” buffers from Fisher Bioreagents have very high purity (assay >99%) and only trace amounts of metal ions, which makes them useful for applications requiring tight control of elemental content.

Technical Tip

CellPURE Biological Buffers are of the highest quality (purity ≥99%). They are tested for low heavy metal content and the absence of endotoxins, nucleases, and proteases. It is important to use only water of the highest quality to prepare the buffer solutions. Water that stands too long in pipes or water produced by a system needing routine maintenance increases the risk of contamination of buffer solutions. Fisher BioReagents provides several grades of high-purity water suitable for the preparation of high-quality buffer solutions.

Electrophoresis

Fisher Bioreagents offers a wide range of reagents for electrophoresis applications. Choosing the right reagents will allow you to achieve optimal results in your experiment. Use our quick selection guides to assist you in finding the right grades of agarose, buffers and ladders.

Compared to polyacrylamide gel, agarose has a lower resolution but wider range of separation. Lower grades of agarose can be contaminated with other polysaccharides, salts and proteins. Such impurities can alter the gelling/melting temperature of agarose solutions or affect the ability to use the recovered nucleic acid sample in a post-electrophoresis application.

Fisher BioReagents offers three different grades of agarose that are functionally tested and pre-qualified for specific applications. Agarose grades for use in electrophoresis of nucleic acids include:

  • Genetic Analysis Grade - Agarose that yields biologically active DNA or RNA. Testing includes enzymatic performance measurements
  • Molecular Biology Grade - Suitable for analytical separation of DNA or RNA
  • PCR Grade - The original agarose for analytical separation of PCR amplicons (<1kb)

Two buffers commonly used for DNA agarose electrophoresis are Tris-acetate with EDTA (TAE; 40mM Tris-acetate, 1mM EDTA) and Tris-borate with EDTA (TBE, 89mM Tris-borate, 2mM EDTA). TAE and TBE have different properties, which makes one more suitable than the other for a specific purpose.
